DspE, an effector of Erwinia amylovora is translocated into plant cells

Abstract

The hrp/dsp gene cluster of Erwinia amylovora is required for pathogenicity in host plants and elicitation of the hypersensitivity response in non-host plants. It encodes structural proteins of a type III secretion system (TTSS), and proteins that are secreted via the TTSS, including two harpins, HrpN and HrpW, putative effectors and regulatory proteins. DspE, which is required for pathogenicity in host plants, is secreted in vitro in hrp gene-inducing media via the TTSS. Using a CyaA reporter system, we demonstrated that DspE is translocated into the cells of Nicotiana tabacum 'Xanthi'. To determine if other proteins from the hrp/dsp gene cluster of E. amylovora are needed for translocation of DspE, we tested translocation in several mutants of E. amylovora. DspE was translocated in both hrpW and hrpK mutants but not in hrpN or hrpJ mutants. These results suggest that HrpN and HrpJ are required for translocation of DspE and may be part of a translocation apparatus in E. amylovora.
