Large volume sample-pretreatment microdevice based on sol-gel 3D matrix

Abstract

Sample-pretreatment is of great interest for micro-total-analysis-system due to the needs of relevant sample concentration and separation techniques for efficient detection. In this study, we demonstrated a novel 3-dimensional sol-gel matrix based microdevice that can process a large volume of the sample and capture target cells with magnetic nanoparticles (MNPs)-mediated colorimetric signal. The antibody-MNP-DNA-Photocleavable linker complexes were immobilized on the sol-gel and the capture was enhanced by 3D matrix and flutter pumping scheme. The concentrated cells were released after UV irradiation, and analyzed based on MNP-mediated colorimetric detection. This novel platform suggests a simple biosensor device with sample-in-answer-out capability.
