Sample-pretreatment of influenza a virus based on the microbead incorporated centrifugal microdevice

Abstract

In this study, we report an advanced sample-pretreatment microdevice that can be applied for RNA purification from nasal swaps of influenza A virus with high speed, sensitivity and reproducibility. We utilized tetraethoxyorthosilicate (TEOS)-treated glass microbeads as an RNA capture matrix, and designed a capillary valve and a siphon channel for the controllable loading of the washing solution and the elution solution, respectively. The PCR cocktail was used as an elution solution to investigate the feasibility of further integration into the PCR chip, and the clinical samples were successfully analyzed. The whole process could be complete in 440 sec, and the RNA capture yield was∼81%.
